Soft Ionization Mass Spectrometry for Studying Noncovalent Interactions

Biochemistry and structural biology have benefited tremendously from the development of electrospray ionization matrix-assisted laser desorption/ionization, two most important soft methods used in modern mass spectrometry. Of importance this context is that noncovalently bound complexes can be maintained gas phase using appropriate sample preparation conditions instrumental settings ESI-MS MALDI-MS. This allows one to determine architecture stoichiometry large noncovalent complexes, binding small molecule ligands cofactors, often even affinities, since unbound states clearly resolved quantified MS. article presents an overview research particular area undertaken by my group at ETH Zurich. I will introduce a number specialized technologies we available our laboratory, including MALDI spectrometry with high-mass detection for measuring molecular weights 100s kDa range; so-called “gap sampler” technology high-throughput experiments; temperature programmed spray source ESI. A selected applications then presented, study GPCRs other membrane proteins their determination melting behavior non-canonical nucleic acid structures, investigation heterogeneous protein protein-based capsids, influence post-translational modifications on profiles.
